- Figure I. Cultured human saphenous veins produce gelatinases MMP-9 and -2 ex vivo. (A) Gelatinolytic bands extracted from cultured veins revealed by gelatin SDS-PAGE zymography. Identity of bands was confirmed by western blotting. (B) Stimulation with PMA (25 nmol/L) and TNF-alpha (10 ng/mL) increased levels of MMP-9 extracted from veins cultured ex vivo for 5 days, showing that they maintain their responsiveness and capacity to produce MMPs.

- Figure II. Ex vivo hemodynamic conditions do not affect levels of MT-MMP-1 in ex vivo perfused veins. Left panel, representative western blot of protein extracts from a matched pair. Right panel, quantification of MT-MMP-1 levels, as determined by densitometric analysis of scanned data, indicating no significant difference in MT-MMP-1 levels between arterial and venous perfusion conditions. P = 0.43; n = 3 independent experiments.
