- Figure I. Expression of LRP in VSMC. Flourescent micrographs of VSMC incubated with 1 µg/mL of monoclonal antibodies against LRP β-subunit (A and B) or α-subunit (C and D). B and D show cell nucleus stained with Hoechst colorant as explained in Methods. A and C, magnification X2000; B and D, magnification X1000. Representative of 3 independent experiments.

- Figure II. Effect of lactoferrin and polyinosinic acid in the binding and internalization of Dil-agLDL by VSMC. VSMC were preincubated for 1 hour at 37°C with medium alone (A) or medium supplemented with lactoferrin (50 µg/mL) (B), or with polyinosinic alone (50 µg/mL) (C). After this period, VSMC were incubated with Dil-agLDL (precipitable fraction; 50 µg/mL) in the presence of the above compounds for 30 minutes at 4°C. Cells were washed and incubated for 4 hours with basal medium. After this period, VSMC were washed, fixed, and photographed (magnification X1000).

- Figure III. Effect of increasing concentrations of anti-LDL receptor antibody on CE content of VSMC incubated with native LDL. VSMC were incubated with native LDL (100 µg/mL) and increasing concentrations of monoclonal antibodies with anti-LDL receptor (12.5, 25, and 50 µg/mL) for 18 hours. VSMCs were exhaustively washed and harvested for measurement of FC and CE. A, TLC showing FC and CE bands corresponding to VSMC incubated with the different antibody concentrations. B, Bar graph showing the quantification of CE bands corresponding to VSMC incubated with increasing concentrations of anti-LDL receptor antibodies (filled bars) and nonimmune IgG (open bars). Results are expressed as percentage of control (CE increase induced by native LDL) and are shown as mean of one experiment performed in triplicate (deviations <5% of the mean do not appear in the computer-orifginated graphs).
